What does mutational burden mean?  

As one goes through from one effective treatment to another to another over time, does one's the mutation burden (different types of mutations???) goes up?

Hi wadvocator,

 

Tumor mutational burden has been defined as "the total number of nonsynonymous mutations present in a tumor" and is emerging as a biomarker which may predict the efficacy of immunotherapy combinations. See "Tumor mutation burden in lung cancer: a new predictive biomarker for immunotherapy or too soon to tell?" 

 

At least with regard to cancers treated with targeted therapies, TMB does appear to increase when resistance develops, as noted here.
